I have the same thing, I think it comes about because we accidentally put more pressure on the right side than on the left.
Looking around a while back I found this:

I have another technique that might be more useful: Lay your palm flat against the right side of your flaccid penis. Now jelq the left side exclusively about 20 times using two fingers from your left hand. Start close to where your penis exits the body. Don't worry about jelqing the entire length of it; without lube it's difficult. Now the left side will (Temporarily!) bulge instead of the right. During a jelq session take a break and do a few of these; I haven't been able to pe for about a year but I believe it would help even everything out.
That being said, my right chamber (The one that's a little bigger) gets a mild, sharp pain sometimes when I masturbate or even when I get an erection. It's not a lot, but I'd much rather it not be there. This started when my right chamber got bigger. You need to correct your grip so you work both sides the same, otherwise you could end up like me. I know people go into autopilot mode when they jelq, and I understand, but seriously pay attention. If you feel anything out of the ordinary: a pop, a rip, or a poke, anything: STOP.
I'm getting back into pe soon but jelqing is out of the question because of what happened to me; pain from jerking off is bad enough and I know pain from jelqing will be much, much worse. Stay safe!
